Изучение структуры генотипов вируса В и определение гетерогенности PrеS1/PrеS2/S-генов на территории Якутии.Методы и материалы. Изучено 16 изолятов, выделенных у больных хроническим гепатитом В из Усть-Алданского района, методом «гнездовой» ПЦР.Результаты. Филогентический анализ нуклеотидных последовательностей показал наличие трех генотипов вируса В – А (44%), С (12%) и D (44%). Проведена амплификация фрагментов генома вируса B соответствующим PreS1/PreS2/S генам методом «гнездовой» ПЦР в сыворотках крови, положительных на ДНК вируса. Анализ нуклеотидных последовательностей изученных изолятов по локализации замен и делеций в том или ином участке PreS1/PreS2/S генов позволил выявить значительное количество замен. Выявлено значительное генетическое многообразие вирусов гепатита В, особенно изолятов, принадлежащих к генотипам ВГВ А и D. Обнаружены аминокислотные замены, обусловливающие способность ВГВ к «диагностическому» ускользанию.Вывод. Обнаружены 3 варианта генотипа вируса В: А, С и D и выявлено генетическое многообразие изолятов генотипа А и D.</p></abstract><trans-abstract xml:lang="en"><p>The purpose of research. Studying of structure of virus B genotypes and definition of PreS1/PreS2/S-genes heterogeneity in territory of Yakutia.Methods and materials. 16 isolates, allocated in patients with chronic hepatitis B from Ust-Aldanskij region by “nested” PCR method, are studied.Results. The phylogenetic analysis of nucleotide sequences has shown presence of three genotypes of a virus B - A (44 %), C (12%) and D 944%). Amplification of virus B genome fragments, corresponding PreS1/PreS2/S-genes by «nested» PCR method in blood serums, positive on DNA virus, is lead. The analysis of nucleotide sequences of studied isolates on localization of replacements and deletions in tHis or that site of PreS1/PreS2/S genes has allowed revealing of a significant amount of replacements. The significant genetic variety of viruses of hepatitis B, especially isolates, belonging to VHB A and D genotypes is revealed.
